Overview

Thyroglobulin is a large glycoprotein produced exclusively by thyroid follicular cells and stored in the colloid of thyroid follicles, where it serves as the scaffold for thyroid hormone synthesis. Its tyrosine residues are iodinated and coupled to generate thyroxine (T4) and triiodothyronine (T3), which are subsequently released into the circulation, making thyroglobulin essential to the production of hormones that govern metabolic rate, growth, and development. Because thyroglobulin is thyroid-specific, measured serum levels function as a clinically valuable tumor marker in the management of differentiated Thyroid Cancer, where, after thyroidectomy and radioactive iodine treatment, persistent or rising concentrations indicate residual or recurrent thyroid tissue. Research relevant to this area centers on the management and surveillance of differentiated Thyroid Cancer treated with surgery and radioactive iodine, the molecular regulation of thyroid cell proliferation including the role of thyroid transcription factor-1, and the spectrum of thyroid tumors encompassing rare and metastatic malignancies. Image-guided ablation techniques for thyroid lesions and broader efforts to improve thyroid care further inform how thyroid pathology is monitored and treated. Interpretation of thyroglobulin measurement depends on assay characteristics and the presence of interfering antibodies. The topic thus connects the protein's biochemical role in hormone synthesis to its central application in detecting and following thyroid malignancy.
